So I'm trying to 100% Assassin's Creed 1. I've noticed that in some of the memory blocks, I have parts greyed out, they are the investigation missions that you do before every assassination. I went back and did the 2nd block (where you complete your first assassination), I did all 6 investigations and then the assassination.
